Phenotyping Data for Suv420h1 (MAQX) Associated with 'immune system' (MP:0005387)

Citrobacter Challenge: Standard Protocol
For each mutant line, 8 female mutant mice plus 8 female wildtype control mice are infected orally with 0.2ml Citrobacter rodentium lux 109 at 6 weeks of age and are monitored over 28 days. Every 2 to 3 days faeces is collected from each mouse and the amount of bacteria in the faeces is enumerated by serial dilutions and plating onto agar plates. Every 3 to 4 days the mice may be imaged under a bioluminescence camera for 5 minutes. At day 14 post infection, 4 mutant and 4 wildtypes are schedule 1 killed. The spleen, liver, caecal patch, caecum, caecal contents and the last 6 cm of colon are extracted and the amount of bacteria in the tissue/contents is enumerated by serial dilution and plating onto agar plates. In addition, the 6 cm of colon is weighed to look for signs of gross hyperplasia, and the last 0.5 cm of most distal colon is removed and fixed for histology. At day 28 post infection the remaining mice are schedule 1 killed and the day 14 post infection procedures repeated. Mice are weighed daily throughout the infection.

Salmonella Challenge: Standard Protocol
For each mutant line, 8 male mutant mice plus 8 male wildtype control mice are infected i.v. with 0.2ml Salmonella Typhimurium TETc at 6 weeks of age and are monitored over 28 days. On day 14 post infection, 4 of the mutant mice and 4 wildtype control mice are schedule 1 killed. The spleen, liver and caecal contents are extracted and bacteria enumerated by serial dilution and plating onto agar plates. In addition 1 lobe of liver and 1/3 of the spleen will be fixed in formaldehyde for histology. On day 28 post infection the remaining mice are bled under terminal anaesthesia via cardiac puncture. Organs are removed and processed in the same way as describe for day 14 post infection. Blood serum is tested for anti TETc specific antibodies by ELISA assay (IgG, IgG1 and IgG2a). Mice are weighed daily throughout the infection.

Plasma Immunoglobulins: Standard Protocol
Plasma Immunoglobulin Assay (Mice were bled between 14 and 16 weeks of age): Non-fasted mice are terminally anaesthetised and blood is collected into Lithium/Heparin coated tubes via the retro-orbital sinus. Plasma is analysed for the following immunoglobulin isotypes (IgG1, IgG2a, IgG2b, IgG3, IgM and IgA) using a multiplex bead array (Luminex).
