NCI-60 Cancer Cell Line Mutation Data

The NCI-60 is a set of 59 human cancer cell lines derived from diverse tissues;
brain, blood and bone marrow, breast, colon, kidney, lung, ovary,
prostate and skin. Since 1992 these cell lines have been subjected to a
battery of experiments including; extensive pharmacological characterisation by
treatment with over 100,000 chemical compounds, chromosome karyotyping and gene
expression analysis using various DNA microarray platforms.
We are sequencing the NCI-60 for mutations in known human cancer genes.
The cell lines were supplied by the NCI/NIH Developmental Therpeutics Program.
The coding exons and immediate flanking intron sequences of selected genes from
the Cancer Gene Census have been PCR amplified and sequenced.
The results of this work have been entered in the COSMIC
database and web site. In addition we have collected other genetic data on these
cell lines that can be viewed through COSMIC.
Further genes are currently being sequenced to expand the mutation data on the NCI-60.

The positive data, DNA sequence mutations, have been confirmed in
at least two independent experiments. We are investigating some putative mutations further
and if real will add them to this list in the future. The absence of a mutation means
the DNA sequence was normal in our hands in the cell line we are using.
